Affomix is developing an integrated strategy for generation and use of the AffomeTM. The Affomix progression path stems from four complementary technology-based approaches:

Y2HEXPRESSTM, a rapid, automated, high-throughput approach for selection of mAbs

  • Target antigen is tested against yeast libraries of all-human mAbs, in the form of single-chain variable region fragments (scFvs);

  • mAb selection by Y2HEXPRESSTM does not require antigen, so it is particularly useful for generating mAbs against proteins that are difficult to produce or purify;  

  • Affomix uses bioinformatics algorithms for prioritizing epitopes to be targeted;

  • mAb selection is carried out using a patent-protected, automated technology;

  • Affomix currently selects mAbs against 150 targets (proteins or selected protein epitopes) per week, which is scalable for the selection of >1000 different targets per week; 

  • All aspects of the selection process are tracked by the Company’s LIMS;

  • With the Affomix Y2HEXPRESSTM service, mAbs are selected in yeast in 6-8 weeks based on affinity and specificity characteristics;

  • The customer is provided with a description of affinity and specificity characteristics, selects desired mAbs and Affomix expresses and purifies the scFvs for the customer;

  • Y2HEXPRESSTM services are available now.


Phage ESCapeTM for mAbs targeting post-translationally modified (PTM) proteins

  • Large numbers of proteins in the proteome are post-translationally modified (e.g., phosphorylation, methylation, glycosylation); such modifications are key events in regulatory processes.  It has proven to be difficult to generate mAbs for characterizing PTM proteins.

  • Affomix is developing Phage ESCapeTM, an automated high-throughput approach to select mAbs that will bind selectively to PTM proteins but not to their unmodified counterparts, or vice versa. 

  • Phage ESCapeTM services will be available soon.


mAb Optimization 

  • Although Affomix selection approaches are designed to generate high quality mAbs in terms of affinity and specificity, depending upon the intended use of the mAbs, refinement might be needed to either increase affinity and/or specificity;

  • Affomix is developing an automated approach to mAb maturation that simultaneously optimizes based on affinity and specificity. 


Protein Quantitation

Affomix is developing new protein quantitation technology. It is being designed for:

  • Protein quantitation in multiplexed mAb arrays

  • Unrivalled sensitivity

  • Expanded dynamic range
